== Modulating CK1δ activity == Due to the fact that CK1δ is involved in regulation of various cellular processes there is high attempts to influence its activity. Since changes of the expression and/or activity as well as the occurrence of mutations within the coding sequence of CK1δ account to the development of various diseases, among them cancer and neurodegenerative diseases like AD, ALS, PD and sleeping disorders, most interest has first concentrated on the development of CK1δ specific small molecule inhibitors (SMIs). Due to the fact, that CK1δ mutants isolated from different tumor entities often exhibit a higher oncogenic potential than wild type CK1δ there are also great efforts to generate SMIs which are more selective inhibiting CK1δ mutants than wild type CK1δ. == The role of specific tissues and organs in transamination == Transamination takes place in several tissues and organs, especially the liver and skeletal muscle, which work together to manage amino groups generated during amino acid catabolism. The liver also contains aspartate aminotransferase. This enzyme catalyzes a unique reaction where oxaloacetate, instead of α-ketoglutarate, serves as the amino-group acceptor. In this reaction, glutamate transfers an amino group to oxaloacetate, forming the amino acid aspartate and regenerating α-ketoglutarate. == Asymptomology == There are documented asymptomatic males who present no ALD symptoms well into their 60s and 70s. It's not understood how they can have an ABCD1 gene variant and possess elevated VLCFAs and not exhibit either Cerebral ALD, Adrenal Insufficiency, or Adrenomyeloneuropathy symptoms. ==== pH ==== Cellular proteins are held in a relatively constant pH in order to prevent changes in the protonation state of amino acids. If the pH drops, some amino acids in the polypeptide chain can become protonated if the pKa of their R groups is higher than the new pH. Protonation can change the charge these R groups have. If the pH raises, some amino acids in the chain can become deprotonated (if the pKa of the R group is lower than the new pH). This also changes the R group charge. Since many amino acids interact with other amino acids based on electrostatic attraction, changing the charge can break these interactions. The loss of these interactions alters the proteins structure, but most importantly it alters the proteins function, which can be beneficial or detrimental. A significant change in pH may even disrupt many interactions the amino acids make and denature (unfold) the protein.
